O6-benzylguanine, a guanine analogue, is an inhibitor of the DNA repair enzyme O6 alkyl guanine DNA alkyl transferase (MGMT/AGT). O6-Benzylguanine acts as an AGT substrate, transferring its benzyl group to the AGT cysteine residue, which irreversibly inactivates AGT and prevents DNA repair. O6-Benzylguanine can induce apoptosis of tumor cells and has antitumor activity.
