Measurement Principle： Under certain conditions, sulfhydryl group will undergo nucleophilic reaction, i.e. sulfhydryl group reacts with 5,5'-dithio-bis-nitrobenzoic acid (DTNB) to produce a yellow color compound with a maximum absorption peak at 412 nm, from which the free sulfhydryl content of protein can be calculated.
